The proceedings, overseen by Judge Lewis Kaplan, are due to begin in New York.
Andrew Brettler, the Duke’s LA-based lead counsel, will present his motion to dismiss the suit. Mr Brettler will argue that by including a reference to "royalty" in her original lawsuit against Jeffrey Epstein, Virginia Roberts Giuffre had made Prince Andrew a potential co-conspirator.
Mr Brettler is expected to argue before Judge Kaplan today that as Ms Giuffre's 2009 agreement with Epstein was made in direct response to the lawsuit, the claim against Prince Andrew - the only member of the royal family to whom it could refer - is invalid.
David Boies, Ms Giuffre’s lawyer, has described the settlement as “irrelevant” to the case.
The Duke is thought unlikely to dial in to the hearing, which is expected to last around an hour.
Judge Kaplan will either make his decision known at the end of the hearing or hand down a written ruling within days, it is thought.
